Homogenates of yeast extract probed with MCA-38F3, MCA-28F2, MCA-31C4, MCA-31F5, MCA-34B12, MCA-38H8 and MCA-32D6. In each case the name of the protein target and the predicted molecular weight in kilodaltons is given at the top of the figure. The Nsr1p protein recognized by MCA-32D6 runs on SDS-PAGE 10-20kDa slower than predicted by its molecular size. The antibodies are mostly specific for a single protein with the exceptions of MCA-31F5 and MCA-32D6. MCA-31F5 recognizes the two closely related proteins Lys20p and Lys21p which are 90% identical at the amino acid level, while MCA-32D6 recognizes not only Nsp1p but also other related nucleoporin proteins which have similar domains (these are referred to as Nups in the figure). However staining with this antibody is specific for nuclear pores in yeast. |
