The antibody is strongly inhibited from binding to AcGFP by the peptide YLLPDNHYLSTQSALSKDNP and to a much lesser degree by the peptide SKDNPEKRDHMIYFGFVTAA. This suggests that the major determinant of antibody binding is SKDNP and sequence N-terminal to this peptide. This region is amino acids 209-213 of GFP and corresponds to a turn between two β-strands of the β barrel GFP structure.
